Effects of different cytokinins on the growth of hope (Humulus lupulus L.) in in vitro culture

This experiment presents comparative analysis of the effect of four different cytokinins, applied to hop explants in three different concentrations, with regard to the growth characteristics û number of shoots, number of nodal segments, the average length of regenerated shoots and root regeneration frequency. The variables cytokinins and concentrations were found to be significantly different. TDZ showed a higher number of regenerated shoots than the other cytokinins, but shoots were deformed and vitrificated with very short internodes. TDZ was found to inhibit root development. The highest root regeneration frequency was obtained from shoots regenerated on media without plant growth regulators (W0Ph). Comparing different concentrations of 2iP, BAP and KIN, results ranged from the average length of regenerated shoots from 18.1 (2 mg/L BAP) to 33.9 (1 mg/L KIN) mm. The respective strength of the different growth regulators in inducing shoot and nodal formation could be classified in decreasing order as TDZ 2iPKINBAP. We found out that in order to obtain whole plants it is the best to cultivate explants on media W0Ph containing KIN at low concentrations or on media W0Ph without growth regulators.
