In vitro fertilization of porcine oocytes: comparison of fertility in AI - boars

The aim of this study was to compare in vitro fertility of highly fertile boars to select those suitable for embryo production and determine conditions for oocyte fertilization in terms of spermatozoa (sp.) treatment and time of gamete coincubation. Four boars, A, B, C and D, were tested. Oocytes were fertilized by sp. isolated by Percoll gradient and preincubated in Tris-buffered medium without or with 1.5 mM caffeine for 30 min. Oocytes were cultured in PYM-3 medium for twelve hours and then fixed, stained with 33258-Hoechst and examined by epifluorescence. The penetration rates were higher for the untreated sp. of A and B boars than for those of C and D boars. The penetration rate increased considerably in D sp. after the caffeine treatment but not in C sp. The normal fertilization rates for untreated sp. were higher in A and B than in C and D boars. They increased slightly in B sp. but decreased in A sp. after the treatment. The 4h and 30 min-coincubation with both treated or untreated A and B sp. was more efficient than the 3h-coincubation interval. It can be concluded that: 1) it is important to test the in vitro fertility of the boars before their use for embryo production; 2) the response of the sp. after their caffeine treatment is variable; 3) the efficiency of fertilization is influenced by the time of gamete coincubation.
