The study of genic variability in Bacillus thuringiensis Brazilian strains for use in the biological control of Plutella xylostella | Estudio de la variabilidad génica en aislados brasileños del Bacillus thuringiensis para la utilización en el control biológico del Plutella xylostella

El objetivo de este trabajo fue el de encontrar diferencias en el contenido de genes cry de los aislados más recientes de Bacillus thuringiensis en relación al linaje HD1, ampliamente utilizado en la formulación de productos comerciales, así como el de evaluar el efecto de los mismos sobre una población de Plutella xylostella, susceptible a la bacteria. Los aislados BR5, BR6, BR11, BR12, BR16, BR17, BR18, BR21, BR37, BR49, BR54, BR75, BR78, BR80 y BR90; además del aislado HD1 y del control negativo B. thuringiensis var. tenebrionis utilizados en este estudio, hacen parte de la colección del Laboratorio de Genética de Bacterias y Biotecnología Aplicada (LGBBA) de la FCAV-UNESP/Jaboticabal-SP. Para la realización del trabajo fueron realizadas reacciones de PCR utilizándose los iniciadores específicos para cry1Aa, cry1Ac, cry1B, cry1C y cry1F. Con las reacciones de PCR fueron observadas las siguientes amplificaciones: HD1 - cry1Aa y B; BR05 - cry 1F; BR06 - cry1Aa, B y Ac; BR11 - cry1B y Ac; BR12 - cry1Aa, B y F; BR16 - cry1B y Ac; BR17 - cry1Aa, Ac y F; BR18 - cry1B y Ac; BR21 - cry1B y Ac; BR37 - cry1Aa, B y F; BR49 - cry1B; BR54 - cry1B, Ac y C; BR75 - cry1Aa, B, C y F; BR78 - cry1B, Ac y F; BR80 - cry1Aa, B y C y BR90 - cry1B y F. En los bioensayos se observó 100% de mortalidad para orugas de P. xylostella con los aislados utilizados, exceptuándose el aislado BR05 que causó 98%, indicando así que para una población susceptible a este insecto, como la utilizada en el presente estudio, la efectividad de la bacteria B. thuringiensis no depende del contenido de genes cry de cada aislado. Sin embargo, se sugiere que los aislados que presentaron perfiles diferentes en relación a los genes cry estudiados, puedan ser empleados en la formulación de nuevos bioformulados, para manejar la resistencia en casos en que los receptores de membrana sean diferentes de los presentados por los insectos resistentes a aislados como el HD-1, comúnmente encontrado en formulaciones comerciales.

The objective of this work was define the differences among cry genes of the more recent brazilian strains of Bacillus thuringiensis in relation to HD1 strain, commonly used in commercial formulations. It was evaluated too its effects on larvae of a susceptible population of Plutella xylostella. The strains BR5, BR6, BR11, BR12, BR16, BR17, BR18, BR21, BR37, BR49, BR54, BR75, BR78, BR80 e BR90 (Brazilian strains), HD1 and negative control (B. thuringiensis var. tenebrionis) used in this study were from "Laboratório de Genética de Bactérias e Biotecnologia Aplicada (LGBBA)" collection, located in the "FCAV-UNESP, Jaboticabal, SP". For accomplishment of this work, the specific starter for cry1Aa had been carried through PCR reactions using itself, cry1Aa, cry1Ac, cry1B, cry1C e cry1F. By the PCR reactions the following amplifications were observed: HD1 - cry1Aa and B; BR05 - cry 1F; BR06 - cry1Aa, B and Ac; BR11 - cry1B and Ac; BR12 - cry1Aa, B and F; BR16 - cry1B and Ac; BR17 - cry1Aa, Ac and F; BR18 - cry1B and Ac; BR21 - cry1B and Ac; BR37 - cry1Aa, B and F; BR49 - cry1B; BR54 - cry1B, Ac and C; BR75 - cry1Aa, B, C and F; BR78 - cry1B, Ac and F; BR80 - cry1Aa, B and C and BR90 - cry1B and F. The bioassays showed mortalities of 100% to P. xylostella larvae by all strains, excepting BR05 that caused 98% of larvae mortality, indicating effectiveness of B. thuringiensis independently of the cry gene present. It is possible to suggest that the strains showing different profiles in relation to the cry genes studied, can be used to new B. thuringiensis formulations trying to manage the resistance in the cases where the membrane receivers are different from HD1 receivers of resistant insects, currently found in B. thuringiensis commercial formulations.
