[Deproteinization process of crab's shells wastes by aspartic proteinase from Mucor pusillus]

Aspartic proteinase from Mucor pussilus culture was isolated and partially purified and was used in deproteinization process of crab's shells wastes. The activity of purified enzyme was 0.624 mmol/ml.min and two percent of enzyme solution was used in deproteinization process. The aim of this research was to optimize the deproteinization of crab's shells by that enzyme. Research was conducted using factorial Randomized Block Design and was carried out in two steps: the first step was aimed to find out the effect of temperature (30 deg. C, 35 deg. C, 40 deg. C and 50 deg. C) and pH (4.5, 5.5 and 6.5); and the second one was aimed to study the effect of substrate concentration (2 percent, 4 percent, 6 percent, 8 percent, 10 percent and 12 percent) and incubation time (2, 4, 8 hours) on Mucor pussilus proteinase activities. The experiment result showed the optimum activity of Mucor pussilus proteinase was at 30 deg. C and 8 hours incubation times. Statistical analysis showed that temperature and incubation time has a significant effect, respectively at a = 0.01 and at a = 0.05 on enzyme activity, while there were no significant effect in pH and substrate concentration, substrate saturation was not detected in addition of substrate up to 12 percent and the increasing of enzyme activity especially at 8 hours incubation followed the increasing substrate concentration up to 12 percent.
