GOX (glucose oxidase) genetic transformation to cumin in order to induce resistance against wilting disease by using Agrobacterium – mediate method

A protocol for the production of transgenic cumin via Agrobacterium mediated genetic transformation was developed. Disarmed Agrobacterium tumifactens EHA 105 and LBA 4404 carring the binary plasmid pBI121 with the nptII gene and GOX gene (glucose oxidase gene) and chitinase gene were evaluated as vector system. A number of parameters were tested with respect to maximizing transformation efficiency, While pre-culture was inhibitory, the bacterial growth phase (optical densityOD600 = (0.6- 0.8), co-cultivation period (2 days), pH of cocutivation medium (5.6) and acetosyringone treatment (100 ىM ) had positive effects on transformation. Different concentrations of kanamycin as seletive (30-100 mg.ml-1) were used for selection of transgenic plants. Plantlets from embryo explants were resistant to kanamycin (average 4%) and translocated to soil.
