Transferring phosphate uptake gene(s) into range legumes in order to enhance the phosphorous uptake.

Phosphorous is one of the essential macronutrients, necessary for growth and development of plants. Adsorption of phosphate on surface of soil colloids and other components, cause the soluble phosphate change to insduble or fixed form. Absorption, translocation, and assimilation of phosphate depends on activity of various plant genes resulting enzymes and other regulatory components. Legume species are able to convert inorganic atmospheric nitrogen to organic compounds via symbiotic rhizobium bacteria. Availability and increase in absorption of phosphate, bring about rapid growth and development of pland, and finaly increase in yield and biomass. Enhancement of uptake as well as efficiency in assimilation of absorbed nutrient in range and forage legumes, facilitates rapid growthi and productivity of the speices. Regarding the importance of phostphate uptake, aim of the present study is to isolate, characterize and tnansfer the phosphate uptake gene(s) to legume plants such as annual medics and lucern, in order to increase the phosphate uptake of transgenic plants. Partial sequence of the inducible phosphate transport gene of Medicago sativa obtanined from GenBank. Two sets of primers designed and approximately 1 kb fragment, similar to that of the MsPT1 fragment obtained, using PCR technique. mRNA from the M. sativa plants, undr stress of P defficiency isolated. However, due to time limitatios, we did not manage to construct the cDNA library. In order to isolate the phophate transporter gene from the lucern, the mutation experiments, using EMS and UV techniques carried out. The mutant seeds planted in both hydroponic and field conditions and M1 population established.
