Development of new diagnostic methods for strawberry virus-free certification [Fragaria x ananassa Duch.; Italy]

Strawberry virus-free certification in Italy, based on plant indexing method, requires the absence of the three main aphid-borne viruses infecting Fragaria spp.: Strawberry mottle virus (SMoV), Strawberry crinkle virus (SCV) and Strawberry mild yellow edge virus (SMYEV). Since grafting leaves to indicator hosts is an expensive and time consuming practice that, in addition, requires greenhouse facility and the serological approaches (except for SMYEV) are unreliable, the possibility to apply an alternative nucleic acid-based detection assay has been investigated. A molecular protocol based on nested RT-PCR was developed to be applied in routine diagnosis. The reliability of this protocol, for SCV, SMYEV and SMoV detection, was compared with the indexing methods, testing 3 plants of 8 different cultivars. Nested RT-PCR allowed to detect all the three viruses in a higher number of plants; 14 out of 24 were positive to virus infection, compared with indexing, which detected the infection only on 6 plants

Sono stati confrontati i risultati dei saggi eseguiti su piante di 8 diverse varietà di fragola per evidenziare la presenza dei 3 virus della fragola maggiormente diffusi nei nostri impianti: Strawberry mottle virus (Virus della maculatura fogliare della fragola = SmoV), Strawberry crinkle virus (Virus dell'arricciamento fogliare della fragola = SCV) e Strawberry mild yellow edge virus (Virus dell'ingiallimento del bordo della fragola = SMYEV), impiegando sia l'indexaggio su indicatori biologici, metodo previsto nei protocolli di certificazione sanitaria, sia la tecnica molecolare nested RT-PCR. Quest'ultimo metodo si è rivelato più sensibile di quello tradizionale; 14 dei 24 campioni saggiati sono infatti risultati infetti dai tre virus sopra menzionati, mentre con l'indexaggio ne risultavano solo 6. La nRT-PCR può perciò essere impiegata nei saggi dei materiali di propagazione, sia per i controlli della certificazione, sia per quelli di quarantena
