Mapping of factors for resistance to leaf-blade feeding by European corn borer (Ostrinia nubilalis) in maize [Zea mays L.; Iowa]

Английский. The European corn borer (ECB), Ostrinia nubilalis Huebner, is an important pest of maize in the US. Breeding for resistance to leaf-feeding by ECB can be more efficient if the genetic basis for resistance is better understood. The objectives of this study were (i) to locate and characterize the genetic factors controlling resistance to leaf blade feeding by ECB larvae in temperate lines and (ii) to compare the locations of the quantitative trait loci (QTLs) detected in the F2:3 and F6:8 generations of the same maize population derived from the cross of inbred lines Mo17 and H99. One hundred and fifty F2:3 lines and the parents and 185 F6:8 lines and the parents were grown in replicated trials in 1989 and 1995, respectively. Plots were infested with ECB larvae and visual leaf blade feeding ratings were obtained for each plot. Lines were genotyped with RFLP and SSR loci and a linkage map was developed. QTL analysis was performed with composite interval mapping. The heritability of family means was 0.77 in the F2:3 generation and 0.91 in the F6:8 generation. The QTLs explained around 50% of the phenotypic variance in both generations. Five QTLs were detected in each generation. A QTL on linkage group 4 near umc123 and a QTL on linkage group 7 near umc110 were detected in both generations. Significant epistatic interactions among QTLs were detected in both generations. Several loci that showed significant interactions did not have significant main effects, indicating that important epistatic interactions will be missed if only interactions between QTLs with significant main effects are tested. Marker-assisted selection will be most efficient when main effects and interaction effects of QTL are included in the selection criteria

Il miglioramento genetico per la resistenza ai danni alle foglie determinati da ECB può essere più efficiente se se ne conosce meglio la base genetica. Gli obiettivi di questa ricerca erano: (i) localizzare e caratterizzare i fattori genetici che controllano la resistenza a ECB nelle linee temperate e (ii) confrontare le localizzazioni dei QTL individuati nelle generazioni F2:3 ed F6:8 della stessa popolazione di mais derivata dall'incrocio fra le linee consanguinee Mo17 e H99. In prove replicate nel 1989 e nel 1995, rispettivamente, sono state allevate 150 linee F2:3 e i parentali e 185 linee F6:8 e i parentali. Le parcelle erano infestate da larve di ECB e per ognuna sono stati valutati quantitativamente i danni alle lamine fogliari riscontrabili visivamente. Le linee sono state genotipizzate con loci RFLP e SSR ed è stata sviluppata una mappa di linkage. L'analisi dei QTL è stata effettuata tramite il composite interval mapping. L'ereditabilità delle medie delle famiglie era di 0,77 nella generazione F2:3 e di 0,91 nella F6:8. I QTL spiegavano circa il 50% della varianza fenotipica in ambedue le generazioni. Sono stati individuati 5 QTL in ogni generazione. In ambedue le generazioni è stato individuato un QTL sul gruppo di linkage 4 vicino a umc123 e un QTL sul gruppo di linkage 7 vicino a umc10. In ambedue le generazioni sono state evidenziate interazioni epistatiche significative fra i QTL. Numerosi loci che presentavano interazioni significative non avevano effetti principali significativi, indicando che possono essere ignorate importanti interazioni epistatiche se sono saggiate solamente le interazioni fra QTL con effetti principali significativi. La selezione assistita da marcatori presenta la massima efficienza quanto sono inseriti nei criteri di selezione gli effetti principali e gli effetti di interazione dei QTL.]
