Callus and multiple shoot production of abaca (Musa Textilis Dee cv. Agbayanon) as influenced by different sources of planting materials and various combinations of synthetic phytohormones

A study was conducted atthe Tissue Culture Laboratory of the Aklan State University from January to September 2004. It aimed to determine the effect of different kinds of planting materials as source of explants and synthetic phytohormone combinations on the callus formation and shoot production of tissue-cultured abaca cv Agbayanon. The study involved two factors: the sources of planting materials designated as Factor A consisting of the following: corm, peeper, and sword leaf sucker; Factor B corresponds to the levels of synthetic phytohormones. The treatments were replicated three times and arranged in a Randomized Complete Block Design. The study showed that the different sources of explants and various combinations of synthetic phytohomrones favorably influenced the callus formation and shoot production of tissue-cultured abaca, Explants taken from sword leaf sucker (65.52 days) and pepper (70.88 days) comparably formed callus earlier than the explants taken from the corm (83.98 days). Likewise, explants derived from the sword leaf sucker and peeper respectively. Moreover, shoots of tissue cultured abaca developed comparable number of leaves with 2.87 for explants taken from sword leaf sucker and 2.82 for peeper. On the other hand, explants growtn in Murashige and Skoog (MS) medium added with 5 parts per million (ppm) benzylaminopurine (BAP) formed callus and shoots earlier than those in the rest of the treatments; the most number of shoots (4.82) were counted from plain MS medium (no phytohormone added). No interaction effect was observed in all the parameters considered in the study.
