Production of chicken antibodies against botulinic toxin A

Traditionally, polyclonal antibodies against botulinic toxin have been prepared from horse or other animals' serum after immunization. In this study we assessed an alternative method namely, the immunization of chickens and extraction of immunoglobulin Y (IgY) from egg yolk. The main advantages of this alternative source of specific antibodies (Ab) are the non invasive antibody sampling and also the cost-effective production. Chicken IgY Ab against botulinic toxin A were raised using Freund's adjuvant. The IgY Ab were extracted from yolk using the so called water dilution method according to Akita and Nakai. The Ab obtained were effective in neutralizing the toxin in a mouse bioassay. Our results show the feasibility to produce egg yolk extracts containing significant immunoneutralizing activity against botulinic toxin A. This offers the possibility of implementing early action oral treatments with specific IgY for person suspected to have ingested contaminated food. Chicken Ab also appear suitable for preparation of low cost immunoreagents
