Changes in indole-acetic acid and zeatin levels during endoreduplication

Changes in endogenous IAA and zeatin level was measured from root tips of Allium cepa to known their role during endoreduplication. Colchicine was used as anti-mitotic agent. Growth parameters viz. length of roots, width of root tips, cell size and nucleus size were compared with the different concentrations of colchicine and distilled water kept as control. DNA content was determined by chromatography. Parallel comparisons were performed to known the influence of endogenous IAA and zeatin levels in endoreduplicated cells. It was observed that zeatin levels and DNA content was higher in colchicine treated cells compared to control. IAA level maintained equal in control and treatment suggests that it has no influence on endoreduplication. Zeatin-IAA ratio was also more in treated cells than in control. The value was more in lower concentration of colchicine. Role of these phytohormones in the process of endoreduplication is discussed.
