An analysis of local epizootic was given for some backyard flocks caused with high pathogenic influenza A virus (HPAI, subtype H5N1) in 8 districts of the Moscow Region. Samples of internal organs (brains, livers, spleens, blood clots) from dead chickens, geese and turkeys were collected not later than 24 h from the moment of poultry death. IV A RNA in field material was indicated by the reverse transcription real-time nest PCR (RT-RT-PCR) and by means of biochips (HA, NA). According to the data from a molecular genetic analysis of strain A/chicken/Moscow/2/2007 all the isolated strains were referred to HPAI/H5N1 of Qinghai-Siberian genotype (QSG). The study of gene homology of the strain A/chicken/Moscow/2/2007 and the HPAI/H5N1 strains isolated in 2005-2007 in different regions of the world has showed that the greatest homology for all the genes was observed with the group of strains isolated in anthropogenic and natural biocenoses in the Black-and-Caspian Sea region in winter 2005-2006 confirmed also by the great number of general amino-acid replacements. The strain A/chicken/Moscow/2/2007 was really different in a molecular genetic aspect from all the other HPAI/H5N1 strains. Twelve unique amino-acid replacements occurred nowhere but in this strain were found in 4 genes of A/chicken/Moscow/2/2007: PB2, PB1, HA and NP. All amino-acid replacements on protein PB1 regarding the consensus HPAI/H5N1 are also unique. This testifies that during 2006 the strain A/chicken/Moscow/2/2007 circulated actively enough. In view of that all the affected backyard poultry was purchased in the market Sadovod in the South-Eastern District of Moscow being the secondary source it is concluded that the epizootic was of manual origin; the primary source of IV A is unknown. The situation like that may be considered as a model when the source of infection is elucidated in cases of potentially possible acts of bioterrorism.
