[Influence of the capacitation conditions on the metabolic activity of spermatozoa treated with freeze-thawing method]

Investigation of changes of the functional status of cryo-preserved bovine spermatozoa after thawing and evaluation of the influence of reduced glutathione and alpha-tocopherol on the metabolic parameters of cells in the conditions of capacitation in vitro was realized. In course of research it was revealed that after spermatozoa incubation in Tyrode's medium in the conditions of 38 deg C there was stated the increasing of respiration intensity, content of adenosine triphosphate, hyperpolarization of the membrane potential of spermatozoa in the dynamics of capacitation in vitro. There was stated the 2-fold increase of oxygen losses from the polarographic cell after 2-hour spermatozoa treatment. In course of 2-hour incubation of thawed spermatozoa treated with alpha-tocopherol (0,05 mM) and with the reduced glutathione (0,5 mM) in the observed suspension there was stated the 1,4-fold and 1,9-fold increased accumulation of adenosine triphosphate, respectively. It was found out that administration of the reduced glutathione (0,5 mM) and alpha-tocopherol (0,05 mM) into a suspension of frozen spermatozoa caused the further respiration activation, increasing of adenosine triphosphate level and increasing of mitochondrion Rd 123 fixation, that proved the perspective possibilities of antioxidant use for the increasing of the functional activity of cryo-preserved bovine spermatozoa
