Evaluation of Resistance of Potato Promising Clones to Main Viral Diseases

In this research, the reaction of sixteen potato-promising clones to PVX, PVA and PVY was evaluated in greenhouse condition. At first, viral infection of each clone was examined using DAS-ELISA. Infected clones were treated by electrotherapy technique to achieve virus free clones. Single nodes of healthy plants were cultured on MS medium. After growing, plantlets were transferred to greenhouse, harvested minitubers about two months later, and used as experimental material. In order to supply inoculum, infected potatoes were collected from Hamedan, Isfahan and Karaj. PVX, PVY and PVA isolates were purified biologically on indicator plants by rub inoculation and replicated on tobacco cv. Samsun. Three weeks old, potato plants were inoculated mechanically with infected tobacco to PVX, PVY and PVA in three independent Randomized Complete Designs (RCD) with three replications and three plantlets in each replicate. After 30 days, the result of DAS-ELISA of each plant was scored. Data analysis of each experiment showed that the reaction of clones to above viruses were significantly different at 1% level. However, the difference among samples in each replicate was not important. Mean comparative of clone absorbs ELISA test indicated that clones 397015-1 and 397015-13 were sensitive to three viruses. Clones 397009-3, 397097-1 and 396151-2 were resistant to PVX and PVY but sensitive to PVA. Clones 397015-2, 397009-6, 397009-8 and 397007-9 were resistant to PVY and PVA but susceptible to PVX. Clone 396151 -8 were resistant PVX but susceptible to PVA and PVY. Clone 397007-14 and 396151-15 were resistant to PVX and PVA but Susceptible to PVY. In addition, clones 397007-11 and 397008-3 were susceptible to PVX and resistant to PVY. Clone with number 397031-2 was susceptible to PVX and PVY. Finally, clone 397008-2 was sensitive to these three viruses.
