Evaluation of Humoral Immunity Response to Immunization with Plasmid Encoding the Anthrax Toxin Protective Antigen (PA) in BALB/c Mice and Sheep.

The protective antigen (PA) of Bacillus anthracis is a potent immunogens and has previously proposed as a vaccine candidate against B. anthracis. PA is an essential component of an effective anthrax vaccine and all anthrax vaccines consist of mainly this antigen. In this study to assess the immune response and protective efficacy of pcDNA3.1+PA plasmid, encoding PA gene of B. anthracis, in mice and sheep, the mentioned plasmid was purified and injected to these animals. Humoral immunity was assessed by ELISA and Total IgG levels were compared in test groups (Sterne vaccine and DNA vaccine) and negative control group. All injections were done intramuscularly by Insulin syringes. Presence of specific PA antibodies was detected in sera of test groups. Challenge results in DNA vaccine group in both animal models showed optimistic protective efficacy of DNA vaccine (over 70%).
