Production of doubled haploid lines as mapping population for grain quality traits in rice

Crosses were between 7 indica and 4 japonica cultivars differing in various quality characteristics such as amylose content, gel consistency, gel temperature and aroma. Out of the 8 F1 hybrids involving indica and japonica crosses being developed, two combinations, PSBR Rc10 x Nipponbare and IR29 x PSBR Rc10, were selected and used in anther culture . The objective of this study was to produce doubled haploid lines to be used as mapping population for grain quality traits in rice. Between these 2 crosses, PSB Rc10 x Nipponbare gave higher value on callus induction (12.63 percent), green plant regeneration (3.82 percent), and the albino regeneration (2.87 percent). A total of 122 green plants were produced and out of these 53.3 percent are spontaneous doubled haploids and 46.7 percent are haploids. Agromorphological, cytological and molecular characterizations using simple sequence repeat (SSR) markers of these DHLs are now being conducted.
