Characterization of Arabidopsis P4H9 T-DNA knockout mutant

Lately, the frequency of major floods has persistently increased. Flooding results in hypoxic or anoxic conditions leading to plant death. Prolyl 4-hydroylase and its relation with oxygen deficiency have been studied in various organisms, but little is known about their role in response to anoxia stress in plants. Arabidopsis thaliana has 13 P4h genes; p4h1 and p4h2 were characterized while little is known about the rest. To this direction, the function of p4h9 gene in A. thaliana was studied by using reverse genetics. Suppression of expression of p4h9 results in shorter roots and hypocotyls and a lower number of root hairs in 10 day-old Arabidopsis seedlings grown under light conditions. However, p4h9 mutant seedlings had longer hypocotyls and roots compared to wild type seedlings grown in the dark. While exposing p4h9 T-DNA mutant plants and wild type plants to anoxic conditions, mutant plants exhibited lower survival rates than the wild type plants indicating a role for Atp4h9 in anoxic stress.
