Antioxidant behaviour of polyphenol mixtures in model systems and correlation with redox potential: distinction of synergetic and/or prooxidant effects

The behaviour of various mixtures of known antioxidants (quercetin, hesperetin, caffeic acid, ascorbic acid and �-carotene), was studied in view of elaborating considerations over the prediction of an eventual prooxidant or synergistic activity. Precisely, measurements were performed by luminol Co(II)-EDTA chemiluminescence, cyclic voltammetry and yeast strain in vivo tests using three different strains; �sod1, �gsh1 and �cta1. The Co(II)-EDTA luminol chemiluminescence tests showed that ascorbic acid mixed with quercetin, hesperetin and catechin has a synergistic effect. On the other hand; ascorbic acid mixed with caffeic acid had a prooxidant activity. The prooxidant activity of ascorbic acid and caffeic acid was well correlated in CV tests. In CV the mixture of ascorbic acid and quercetin showed a synergistic effect, which also correlated well in CL. Due to the low solubility of �-carotene in the media used, this compound was studied by in-vivo tests only. Cells were treated with H2O2 in an identical manner, as above. Cells supplemented with 50�M �-carotene did better than H2O2-treated �sod1 cells (15% at 12 hours), but at 5�� grew slightly less. Ascorbic acid supplementation had no effect on �cta1 cells at any concentration.
