Development of SCAR markers linked to resistance to Fusarium oxysporum f. sp. cucumerinum in cucumber (Cucumis sativus L.)

In cucumber, resistance to fungal rot induced by the pathogen Fusarium oxysporum formae specialis cucumerinum (FOC) appears to be under the control of a single dominant gene (Foc). In an earlier stage of this project, four putative markers - linked to Foc - were identified following bulked segregant analysis and by determining marker segregation among individuals comprising the bulks. In the framework of the present study, the linkage of these markers was assessed employing the entirety of the F2 mapping family. It was concluded that only one AFLP marker (E12M5O) maintained linkage to Foc (in coupling phase) throughout the entire progeny level. Since AFLP markers are not adapted for large-scale marker-assisted selection, our second objective was to de a fast, cheap and reliable PCR-based assay. Towards this goal, we employed AFLP-mediated mini-sequencing technology to reduce the complexity of the AFLP pattern and to ensure sequencing of the truly linked AFLP band. The amended AFLP marker (El2M5O+ T(134)) maps at a distance of 6 cM from the resistance locus Foc, and was successfully converted into sequence-characterized amplified region markers (SCAR). Two dominant SCAR markers were generated from the original AFLP counterpart. SCAR marker SCEI2M5OA sub(114) tagged each allele with a different size marker while SCEl2M5OB sub(113) produced only one fragment linked with the dominant allele Foc.
