Bacterial canker and wilt of tomato plants have been recurrently observed in all major tomato-growing regions in Serbia over past several years. Samples of diseased tomato plants were therefore collected from different tomato-growing regions in order to identify the causal agent using conventional identification methods, and to confirm the identity of isolated bacterial strains using molecular and serological methods. All of the 60 isolated strains were identified as Clavibacter michiganensis subsp. michiganensis. A comparison of seed extraction procedures for the detection of C. michiganensis subsp. michiganensis in infested tomato seed lots showed that the extraction method that included grinding and soaking in PBS buffer was significantly better at detecting the pathogen than the method involving only soaking in PBS. A simultaneous comparison of the sensitivities of five methods for the detection of C. michiganensis subsp. michiganensis in order to determine bacterium detection threshold in tomato seed lots showed that in samples of 5000 seeds containing one infested seed, C. michiganensis subsp. michiganensis could be detected only by Bio-PCR and agar plating assay on three semi-selective media (mSCM, D2ANX and mCNS), while negative results were obtained by ELISA test, and direct and Enrichment PCR. A high correlation was found between disease incidence and infested seeds. Investigation of the host range of C. michiganensis subsp. michiganensis strains originating from Serbia confirmed tomato and pepper as the baterium hosts. The effect of bacteriocides and a plant activator in terms of a reduction of population sizes of C. michiganensis subsp. michiganensis in the greenhouse were investigated, too.
