Differential proteomics analysis of leaf development at rice (oryza safiva) seedling stage

Китайский. 【目的】从蛋白质组学角度对杂交水稻汕优63苗期的叶片蛋白质组变化进行研究,以揭示水稻苗期叶片蛋白质组的表达差异。【方法】蛋白质点经双向电泳得到分离,并采用Imagemaster 2D Elite5.0软件对所得到的双向电泳图谱进行分析比较。【结果】经图谱分析共得到差异蛋白质点41个,其中三叶期后新增蛋白质点17个,包括9个仅在三叶期出现特异的特异蛋白质点,13个蛋白质点表达丰度先逐渐增强随后减弱或在消失,其它11个蛋白质点中,自二叶期开始表达丰度呈现下降的蛋白质点有3个,保持上升的有6个,不规律出现的有2个;其中的15个蛋白质点经质谱分析得到鉴定。【结论】蛋白质是细胞功能执行者,受细胞功能或细胞环境变化的影响,因此,上述蛋白质发生变化的原因可能与水稻苗期分蘖发生有关系。

[ Objective ] In order to understand the function of differential expression proteins in leaves of seedlings, the proteins of leaves at the seedling stage of hybrid Shanyou63 were studied by using approach of plant proteomics. [ Method ] Proteins were isolated with 2-DE and then analyzed with Imagemaster 2D Elite 5.0. [Result] The result showed that the number of detected differential expression protein spots was 41, among which there were 17 new protein spots that appeared after the 3-leaf stages including 9 special protein spots which only appeared at 3-leaf stages. Thirteen protein spots increased first and then decreased gradually or disappeared in expression abundance. There were 3 protein spots decreased, 6 protein spots increased in expression abundance and 2 protein spots appeared irregularly among the other 11 protein spots after 2-leaf stage, andl5 protein spots were identified by ESI-Q MS/MS. [Conclusion] The protein is the performer of cell's function and influenced by the variation of cell's function or cell environment, therefore, the reason that caused the above protein changes should be related with the occurrence of tillering at rice seedling stage.
