Correlation between polymorphism of GARS-AIRS-GART gene coding region and IMP contents in chickens

Китайский. 以鸡GAR-AIR-GART基因为侯选基因,隐性白羽鸡、丝羽乌骨鸡、萧山鸡、白耳鸡、藏鸡为实验材料,采用PCR-SSCP法对GAR-AIR-GART基因所有外显子进行SNPs检测.结果在外显子3、4和21中各发现了一个单核苷酸多态性位点.方差分析结果表明:外显子3和外显子21的基因型与IMP含量之间不存在显著关联.夕\}显子4中的6545处的C→A突变位点对胸肌IMP含量的影响在不同鸡种之间的表现不完全一致.在隐性白羽鸡、萧山鸡和白耳鸡群体中.三种基因型个体之间的IMP含量差异均不显著.在丝羽乌骨鸡和藏鸡群体中,AA型个体的IMP含量显著高于AC型和CC型,推测该位点的基因型效应可能是其等位基因与丝羽鸟骨鸡、藏鸡群体中的特殊遗传背景之间存在交互作用。或者是与影响肌肉IMP含量的QTL之间存在着暂时的连锁不平衡的缘故.[著者文摘]

This study was designed to investigate the single nucleotide polymorphism (SNP) of GARS-AIRS-GART gene in various breeds,including Recessive White chickens, Silkies chickens,Baier chickens, Tibetan chickens and Xiaoshan chickens. The primers for all the 21 exons in GARS-AIRS-GART gene were designed and the SNPs were detected by PCR-SSCP and DNA sequencing. Three SNPs were detected in the exon 3,exon 4 and exon 21. The results from analysis of variation suggested that there was no significant relationship between exon 3 and exon 21 SNP sites and IMP content in chickens,the effect of exon 4 polymorphic site on IMP content of breast muscle were not consistent in different chicken breeds. No difference of IMP content was found among CC, AC and AA individuals in RW, XS and BE populations. In SC and TC,IMP content of individuals with AA genotype was significantly higher than that of individuals with AC and CC genotype. We argued that the genotype effects of this locus may be an interaction between its allele and the specific backgrounds of Silkies and Tibetan chickens,or a temporary linkage disequilibrium between the locus and the QTL which affects IMP contents of muscle in chickens.
