[Use of molecular markers to select and analyze the quantitative traits of grain and forage maize in the humid zones from Spain] | Utilización de marcadores moleculares para la selección y disección de caracteres cuantitativos de maíz grano y forrajero en las zonas húmedas de España

Испанский язык; кастильский. El Centro de Investigaciones Agrarias de Mabegondo (CIAM) posee un programa de mejora de maíz que se está desarrollando desde hace 30 años. Hasta ahora la mejora se ha realizado únicamente mediante los métodos tradicionales, lo que implica un trabajo de entre 8 a 10 años para la obtención de una línea o variedad mejorada y aún pasado este tiempo, no se puede asegurar que esa línea sea totalmente pura, molecularmente hablando. Es por ello, que para tratar de acelerar este proceso, se ha planteado la inclusión en los programas de mejora de herramientas moleculares como son los microsatélites (SSR) para la búsqueda de QTLs (Quantitative Trait Loci), que a su vez servirán para la aplicación de una selección asistida por marcadores (MAS), siendo este el objetivo principal y final de esta tesis doctoral. Se emplearon como parentales las líneas EC136 y EC151, obtenidas en el CIAM que, a priori, mostraban buena aptitud combinatoria y sus características agronómicas a estudio eran lo suficientemente diferentes para poder mapear los QTLs asociados a estas. Para los estudios de mapeo se empleó la generación F2 derivada del cruce de ambos parentales. Como caracteres de interés se evaluaron el Rendimiento Grano, Rendimiento Forrajero, Encamado, Floración Femenina, Vigor Temprano, contenido en Proteína Bruta, contenido en Almidón, contenido en Fibra Ácido-Detergente, la Digestibilidad de la Materia Orgánica in vitro y la resistencia al hongo Fusarium graminearum Schw. Английский. Maize breeding have traditionally involved the selection of better varieties using phenotypic characterization. Despite historical success the traditional methods are time consuming and do not assure a pure inbred line. The advent of molecular genetic tools such as molecular markers has enabled accelerated the selection of better lines and varieties, these molecular tools are very different and their selection depends on the main objectives. Microsatellites or Simple Secuence Repeats (SSRs) represent one of most useful category of molecular markers. They have the advantage of being efficient, stable, co-dominant, abundant and distributed throughout the genome hence. Owing to the above advantages SSR markers were chosen as marker systems for the study. The principal objective in this study was to map the QTLs (Quantitative Trait Loci) associated with agronomic traits of interest. The linked markers can then be used for Marker Assisted Selection (MAS) to devise an accelerated breeding strategy for traits of agronomic interest in maize. Maize inbred lines EC136 and EC151 obtained in previous CIAM's breeding programs were used throughout this study. Although belonging to same heterotic group these inbred lines are reasonably heterogeneous hence, uses as parents in this study. Consistently, they agronomic phenotypes are different and therefore might have some genome wide differences that might be linked with the observed differences. The targeted agronomic traits include Grain Yield (GY), Forage yield (FY), Early Vigour (EV), Lodging (Lo), Flowering (Fl), Protein content (PC), Starch content (SC), Detergent-Acid Fibre content (DAF), in vitro Organic Material Digestibility (IVOMD) and Fusarium graminearum Schw. resistance (FGR).
