Asexual regeneration of two species of Eucalyptus grandis and Eucalyptus globulus by tissue culture.

Eucalyptus grandis and Eucalyptus globulus are important industrial value fast growing trees. These are cultivated for their wood fibers which are widely used in the paper industry, domestic fuel and charchoal production. This study was carried out as a step toward clonal propagation of superior genotypes. Apical and axillary buds from adult elite trees in forests of north of Iran was collected in different seasons. Immersion buds in 70% ethanol solution for 30 sec and subsequently application of 0.1 % mercuric chloride solution in 7 min in summer is the best treatment for surface sterilization. These explants were cultured on a modified MS medium containing half strength Nitrate and cytokinines (BAP, 2iP, Kinetin, Zeatin, GA3,TDZ) in (0 -1 mg l-1) and auxines (IBA, IAA) in (0.1-0.01 mgl-1 ).The rate of shoot multiplication, shoot length growth and other responses were studied. All of them showed rapid multiplication, when cultured on MS medium with 0.1 BAP, 0.2 Kin, 0.1 GA, 0.01 mgl-1 IBA . Shoots from the multiplication medium was transferred in MS medium (half strength Nitrate) containing 1 mgl-1 Zeatin, 0.1 mgl-1 IAA for shoot elongation. Elongated shoots excised at 1.5 cm in length were rooted in 1/2 MS with 0.5 mgl-1 IBA plus NAA. These plants were established successfully in the greenhouse conditions.
