A system is described for the separation and purification of the protein and lipoprotein components of biologicqi tissues and fluids and is applied to the fractionation of human plasma. Simultaneous control of all the variables of a multi - component system permits the choice of the optimum conditions for the separation of a component from a mixture. the separation reported have thus for been achieved by the precise control of five variables: 1) salt 2) protein 3) water miscible organic precipitant 4) PH 5) temperature. Concentration of salt is maintained as low as possible in order to take advantage of the diversity of protein - electrolyte interactions to avoid the necessary of diaZvsis. Concentration of the organic precipitant - ethanol in this study and the temperature are maintained as low as passible in order to minimize protein denaturation. The temperature has been maintained between 0 and the freezing point of the solution. the PH has been controlled by buffering with acetates or other buffers of known ionic strength.
