Promotion of flowering by the chilling treatment of Limonium sinuatum Mill. in tissue culture

We were developed the chilling treatment in tissue culture for Limonium sinuatum Mill. that came into flower before the end of the year. First we cultured meristem of Limonium using Murashige and Skoog (MS) medium added 0.1 mg/l BA, 30.0 g/l sucrose and 8.0 g/l ager and controlled pH 5.8. Its meristem cultured plantlets were propagated by MS medium added 0.1 mg/l BA, 30.0 g/l sucrose and 8.0 g/l ager and controlled pH 5.8. It was a better medium taking root that used Hyponex medium added 5.0 g/l Hyponex and 0.1 mg/l NAA and controlled pH 5.8. We used a plastic case to be length 193 mm x width 115 mm x height 70 mm square that was opened 6 circles 9.0 mm across and covered by fluoro carbon membrane. Also we used vermiculite to a culture medium poured above Hyponex medium and planted a cutting of the cultured plantlets becoming wedge on it's base. After rooting culture these plastic cases were vernalized in 4degC under lighted for 30 days at 500 lx, 16 hr. In result it was possible to come into flower before the end of the year. To bring up seedlings after potted, the cultivated seedlings into air-condition plastic house were better than the cultivated seedlings into air-condition plastic house nothing but night. At October, it's bolting rate became at 100%. As a result of examined cutting flowers, it seemed all the same. To the seedlings to vernalized and cultivated into air-condition plastic house, it was harvested 4.9 stems/plant in November and 9.2 stems/plant until March. To cultivated time of seedlings after potting, 30 days seedlings were compared with 20 days seedlings and 10 days seedlings. As a result in November 30 days seedlings were harvested 6.5 stems/plant most and better than 20 days seedling or 10 days seedlings. 20 days seedlings had second place. However at a number of flowers before the end of the year and until March, 30 days seedlings were as same as 20 days seedlings.
