Molecular characterization of cucurbit aphid-borne yellows virus isolates associated with 'namamarako' in the Philippines and 'Mara ba' in Thailand

Two isolates of cucurbit aphid-borne yellows virus (CABYV) associated with 'Namamarako' (NMK) in the Philippines and 'Mara Ba' in Thailand were characterized. Symptoms on bittergourd leaf infected with MBV are similar to the symptoms of NMK infection, which is characterized by interveinal chlorosis, green vein bonding, wrinkling and thickening. However, serological analysis of symptomatic plants by DAS-ELISA revealed that NMK and MBV are not serologically related as indicated by weak positive reactions of MBV-infected plants against NMK antisera. Further characterization of independent isolates by RT-PCR using primer pair 5'-GAATACGGTCGCGGCTAGAAATC-3' and 5'-CTATTTCGGGTTC. TGGACCTGGC-3', designed to amplify the open reading frame 4 (coat protein gene) of CABYV revealed a single DNA product of about 600 base pairs. Cloning and sequencing of the 600 bp RT-PCR product from both isolates revealed 81% homology between the two isolates based on amino acid sequence, indicating that NMK and MBV are two different isolates of CABYV. Analysis of the coat protein sequences indicated that NMK is 95% identical with CABYV, which was first reported in France and is closely related to CABYV isolates in Italy, Spain and Tunisia. MBV on the other hand is 81% identical with CABYV in France and is closely related to CABYV isolates in China.
