Identificación del material de recepción del Banco de Germoplasma de Olivo de Córdoba mediante marcadores SSRs
2010
Amokrane, K.
In this work, a set of 14 SSR markers has been selected for the study of 221 Olea europaea L. plants belonging to 74 accessions from the reception material of the World Germplasm Bank of Olive (BGMO). These markers have been very effective, showing high values in the estimated genetic diversity indices such as total number of alleles, levels of observed and expected heterozygosity and polymorphic information content, indicating that the analyzed collection has a high genetic richness. In this sense, it was remarkable the presence of 41 unique alleles and 21 alleles not previously described in the varieties currently included in the BGMO. The analysis of the 14 SSR molecular profiles allowed the identification of 102 different genotypes in the 221 plants analyzed. This high number of genotypes was due to the presence of plants of different genotypes within an accession. In fact, in 17 accessions and in five accessions tow and three different genotypes were detected, respectively. These cases may be due to errors during the process of labelling and shipping in the country of origin and propagation, labelling and identification errors of the samples in BGMO. The comparison of molecular profiles also allowed the identification of seven duplicate genotypes with cultivars currently conserved in the BGMO. The availability of endocarp in 16 accessions allowed their morphological analysis which confirmed the identification made by SSR markers. The analysis of genetic relationships among the 102 different genotypes confirmed the great genetic variability of this collection and pointed out a clear grouping of accessions from Syria
