The present study was carried out to investigate possible effects of lead acetate administration on porcine ovarian granulosa cells in relation to insulin–like growth factor I (IGF–I) and progesterone (P4) release. Ovarian granulosa cells were incubated with/without lead acetate for 18 hours: group Max (0.5 mg Pb per ml), group A (1:1 dilution), group B (1:5), group C (1:7), group D (1:10) and the control group without Pb addition and these substances were assessed by RIA. Lead was shown to be capable of accumulation in porcine ovarian granulosa cells. Obtained data indicate Pb–induced inhibition of IGF–I release at lower concentrations by ovarian granulosa cells. However, Pb administration did not show any such effect as no differences were observed between the experimental groups and control in regards to release of P4. These observations suggest possible involvement of lead, as a heavy metal and risk factor of environment, on release of hormonal substances by porcine ovarian cells. Our observations represent the demonstration influence of Pb on secretory activity of porcine ovarian granulosa cells.
