[目的]调查家蚕幼虫期类胡萝卜素水平的变化及cbp基因的mRNA表达特征。[方法]选取黄茧和绿茧品种家蚕3、4和5龄期幼虫的丝腺,参照Takara Trizol plus试剂说明提取总RNA,根据文献提供的cbp基因序列设计特异引物,采用RT-PCR方法调查不同茧色品种家蚕幼虫不同龄期丝腺中cbp基因mRNA的表达特征和主要组织中类胡萝卜素的水平变化。[结果]cbp基因的mRNA表达,在3和5龄期各品种家蚕均维持一定水平,但在4龄期表达量很低或不表达。同一组织的紫外可见光谱扫描结果显示,4龄期家蚕丝腺类胡萝卜素的含量极低,不同茧色品种cbp基因的mRNA表达也存在差异,绿茧品种仅表达1个缺失第2外显子的转录本,而所有黄色茧品种还可表达1个序列完整的转录本。[结论]家蚕体内类胡萝卜素的水平与cbp基因mRNA表达相关。_
Показать больше [+] Меньше [-]Английский. [Objective] The study aimed to investigate the carotenoid level change and cbp gene expression character in silkworm larvae. [Method]The silk glands of 3th,4th,5th instar larvae of silkworm with yellow and green cocoon strains were selected to extract their total DNA by referring to the reagent introduction of Takara Trizol plus and the special primer was designed on base of cbp gene sequence provided by the literature. The mRNA expression character of cbp gene and the change of carotenoid level in the main tissue of silkworm larvae with different colored cocoon strains at different instars was investigated by RT-PCR method. [Result]The mRNA expression of cbp gene could keep some level in 3th and 5th instar larvae of different silkworm strains and was almost undetectable in 4th instar larvae. The UV-Vis spectrum study on the carotenoid level in the same tissue of silkworm showed that the carotenoid content in the silk glands of 4th instar silkworm was very low. The mRNA expressions of cbp gene were different among different colored cocoon strains. Only one mRNA transcript lacking exon 2 was found in green cocoon strains and a complete mRNA transcript was expressed in all yellow cocoon strains. [Conclusion] The mRNA expression of cbp gene was correlated to the carotenoid level in silkworm._
