RFLP analysis of the ribosomal DNA ITS Region of three geographical populations of Tegillarca granosa | 3个地理居群泥蚶核糖体DNA ITS区的RFLP分析

RFLP analysis was used to investigate the genetic diversity and phylogenetic relationship of three populations of Tegillarca granosa based on the ribosomal internal transcribed spacer (ITS) region. The results showed that BstHH I, Hae III and Sse9 I were polymorphic restriction endonucleases in PCR product digestion, while Msp I, Rsa I and Taq I were monomorphic. Nucleotide diversity index (π value) of population in Ningbo was 0.0017, and π values in Wenzhou and Fujian were 0.0012 and 0.0011, respectively. It was shown that the genetic diversity was the highest in Ningbo population followed by Wenzhou population and Ningbo population. According to the Pnet value among three populations and cluster analysis results, it was concluded that the phylogenetic relationship between Fujian population and Wenzhou population was farther than that of Fujian population and Ningbo population and the geography relationship of Wenzhou population and Ningbo population was the closest.
