Genetic diversity among some sudanese sorghum accessions using molecular markers and phenotypic characterization

The aim of the present study was to assess genetic diversity among a set of 95 newly collected Sudanese sorghum accessions. These accessions were provided by the Plant Genetic Resources Unit (PGRU) of the Agricultural Research Corporation (ARC) Wad Medani, Sudan. The field experiment was done at Gezira Research Farm in Wad Medani, in 2008 season. Sorghum accessions were morpho-agronomically characterized in the field using the International Board for Plant Genetic Resources (IBPGR) Descriptor list for sorghum (IBPGR, 1984) and Distinctiveness, Uniformity and Stability (DUS)-test guidelines for sorghum (ICRISAT). The agronomic performance of the accessions was evaluated in, regular field experiments with two replications. Representative sample of 48 accessions based on morphological evaluation was used for Simple Sequence Repeats (SSR) markers analysis using a set of 13 SSR markers. A highly significant difference among accessions for all of the traits was revealed. Days to flowering, plant height, growth rate, panicle weight, panicle length, total biomass, 1000 grain weight and grain yield/plant showed wide variation, while stem diameter showed a narrower range of variation. The grain yield/plant was positively correlated to Panicle weight. The result showed different distribution frequencies of morphological classes of panicle compactness and shape. Based on their morphological characters the 95 accessions were grouped into four main groups scattered to 13 cluster groups. The number of accessions per cluster ranged between 2 to 14. The SSR markers generated 152 alleles, the number of alleles revealed by each locus ranged from 8 to 15 with an average of 11.7 A wide range of fragement size was observed from 147 bp to 388 bp
