Comparative study on effectiveness of using various PCR-markers for X-virus resistant potato (Solanum tuberosum) genotype selection was realized in the Republic of Belarus. Screening of potato breeding and genetic material for potato X-virus resistance using CAPS markers IPM4, CP60 and allele-specific marker 221R was done. Occurrence of virus in leaf material was determined by enzyme immunoassay sandwich method; for allele-specific marker 221R assessment there was used Heldak technique; for amplification reaction to IPM4 marker there was chosen Bendahmane technique optimized for specific condition by article authors; occurrence of CP60 marker was determined by updated Gebhardt et al. technique. Comparative analysis of the efficiency of PCR-markers revealed CP60, IMP4 and 221R for the selection of reistant to X-virus potato genotypes showed that CAPS marker IPM4 in combination with allele-specific marker 221R provided exact and efficient molecular diagnostic of potato X-virus resistance. There were revealed 13 genotype cultivars (varieties Heidrun, Assia, Resurs, Delfin, Neptun, Odissei, Zhuravinka, Darwina, Sante, Dubrava, Olga, Binella, Uladar) which were carrying gene Rx (adg) with both marker alleles (IPM4 and 221R) and expressed high X-virus resistance of potato in the field environment conditions.
