Effects of mediums on the growth of Salvia miltiorrhiza hairy roots and accumulation of tanshinones in S.miltiorrhiza | 培养基中不同营养元素对丹参毛状根生长及丹参酮类积累的影响

Китайский. 研究6,7_V和MS培养基中不同营养元素对丹参毛状根生长及丹参酮类成分积累的影响,探求更有效提高丹参毛状根中丹参酮含量的方法。以6,7-V培养基为基本培养基,分别将MS培养基的大量、微量和有机元素替换6,7-V培养基的相应成分,取培养21 d的毛状根测定其鲜质量,用高效液相色谱(HPLC)法测定培养基和毛状根中二氢丹参酮Ⅰ、丹参酮Ⅰ、丹参酮ⅡA的含量;以6,7-V培养基为基本培养基,以其大量元素为基础,分别用MS培养基中的大量元素KNO3、NH4NO3、MgSO4•7H2O、K2HPO4替换6,7-V培养基的相应大量元素组分,取培养21 d的毛状根测定其鲜质量,利用HPLC法测定培养基和毛状根中的二氢丹参酮Ⅰ、丹参酮Ⅰ、丹参酮ⅡA含量,分析各种培养基对丹参毛状根生长及丹参酮类成分积累的影响。MS培养基的大量元素可显著促进丹参酮类成分的积累,但不利于丹参毛状根的生长,微量元素和有机元素对毛状根的生长和丹参酮类成分的积累无显著影响。以MS培养基的大量元素K2HPO4替换6,7-V培养基的大量元素NaH2PO4•H2O和Na2HPO4时,培养基中丹参酮ⅡA含量最低;以MS培养基的大量元素MgSO4•7H2O替换6,7-V培养基的大量元素_MgSO4•7H2O时,丹参毛状根生长速度最快,且培养基中的丹参酮ⅡA含量高于其他相应处理;以MS培养基的大量元素NH4NO3替换6,7-V培养基的大量元素(NH4)2SO4时,培养基中的二氢丹参酮Ⅰ、丹参酮Ⅰ含量均为最低,同时毛状根中丹参酮ⅡA的含量低于其他相应处理。丹参毛状根培养基中的大量元素显著影响毛状根的生长和有效成分的积累;减少MS培养基的大量元素MgSO4•7H2O的含量,可以促进毛状根的生长和丹参酮ⅡA的释放;而大量元素中含量过多的NH4NO3不利于丹参毛状根的生长,同时也不利于丹参酮类成分的积累和释放。

The research was to study the effect of different nutrient elements in 6,7-V liquid medium and MS liquid medium on the growth and accumulation of tanshinones in hairy root of Salvia miltiorrhiza, and to enhance the tanshinones content in S.miltiorrhiza hairy root effectively. With 6,7-V medium as the basic culture medium, taking MS medium’s macroelement, microelement and organic element to replace the corresponding ingredient of 6, 7-V medium, the growth and contents of dihydrotanshinoneⅠ, tanshinoneⅠ, and tanshinoneⅡA were determined by high performance liquid chromatography(HPLC) method after cultivating 21 d; Taking 6, 7-V medium as the basic culture medium, and its macroelement as the foundation, and MS medium macroelement’s KNO3, NH4NO3, MgSO4•7H2O, K2HPO4 to replace the corresponding ingredient of the 6,7-V medium,the effects of different mediums on the growth and accumulation of tanshinones in hairy root of S.miltiorrhiza were analyzed. MS liquid medium’s macroelement may obviously promote accumulation of tanshinones, but hold back the growth of hairy root. Microelement and organic element can not affect the growth and accumulation of tanshinones significantly.When MS medium macroelement’s K2HPO4 replaces 6,7_V medium NaH_2PO4•H2O and Na2HPO4, tanshinoneⅡA content is the lowest in medium;When MS medium macroelement’sMgSO4•7H2O replaces 6,7-V medium MgSO4•7H2O, the hairy root grows most quickly,and tanshinoneⅡA content is the highest in medium; When MS medium macroelement’s NH4NO3 replaces the 6,7-V culture medium (NH4)2SO4, the contents of dihydrotanshinone and tanshinoneⅠare the lowest in the medium, simultaneously tanshinoneⅡA content is lower than that of other four groups in the hairy root. Macroelement affects the growth and concentration of tanshinones significantly; Reducing the content of MgSO4•7H2O in the MS medium may promote the growth of hairy root and the release of tanshinoneⅡA; However, in the macroelements excessive NH4NO3 does not favor the growth of hairy root, the accumulation and release of tanshinones.
