Today using artificial reproduction methods in livestock reproduction has been considered, especially for the breeds that are in danger of extinction and Caspian horse is one of these considered races in this regard as well. The artificial insemination technique is the one of the first methods of artificial reproduction. In order to find the best diluent for keeping sperm of Caspian stallions in fresh and frozen conditions for using in artificial insemination, 7 Caspian stallions were used in this plan. After collecting sperm from stallions by jumping and artificial vagina method and getting its gel, the sperm was mixed with one of this four diluent solutions including solution number one: 2 gr powdered milk without fat + 4.9 gr glucose + 15,000 IU penicillin and 150,000 µg streptomycin and 92 ml sterile distiled water (pH of solution = 6.7-7.2), solution number two: only sterilized and homogenized milk, solution number three: 3.4 gr trace + 5 gr fructose + citric acid 1.99 gr + 14 ml egg yolk + 100cc sterile distiled water + 150,000 IU penicillin and 150,000 µg streptomycin and solution number 4 containing standard diluent of bull sperm and was studied in different times for checking quality Finally it was determined that sterilized and homogenized milk as diluent has shown better response.
