Identification and frequency estimation of Scrapie resistant genotypes among Iranian sheep populations using Realtime PCR

Scrapie is a neurodegenerative disease in sheep caused by proteinaceous particles (prions) devoid of nucleic acid and susceptibility to that is controlled mainly by point mutations at the PRNP locus particularly the three codons 136, 154 and 171. A sample of Iranian sheep population including 120 individuals from eight populations (Mehraban, Moghani, Sanjabi, Kurdi Khorasan, Kurdi Kurdistan, Kaboud-e-Shiraz, Zandi, and Karakul) were genotyped in three codon using TaqMan RealTime PCR. Some genotypes were directly sequenced in order to confirming results. Allele and genotype frequencies were obtained by direct counting. Four genotyping methodes including present one, PCR-RFLP, direct sequencing and RealTime melting curve analysis were finaly comparisoned on accuracy, cost, facilities and some criteria. These frequencies had no significantly differences (P0.05) both among sampled populations and within total population. Due to differences in the type of allele and genotype between present and previous studies and the lack of significant difference, analysis and evaluation of susceptibility to Scrapie in the studied population was not possible based on previous documentations. So it is might that SNP affected on susceptibility to scrapie in Iranian sheep other than previously reported ones. To finding them, it is needed to infected cases or experimental scrapie. Comparison of genotyping methods showed that TaqMan RealTime PCR has general efficiency higher than other comparisoned ones. Therfore, it could be probably presented in market as a molecular diagnostic kit.
