Effects of culture media on the growth of seedlings derived from embryo culture in Paphiopedilum philippinense (Tropical orchid).

Small portions of immature embryos were cultured on half strength MS basal medium, supplemented with 0.5 mg/l of naphthalene acetic acid (NAA) and 0.1 mg/l of 6-benzylaminopurine (BAP), containing 15% (V/V) coconut water, 20 g/l sucrose and 8 g/l difco-bacto agar. After eight months, protocorms with unrooted shoots were tested to various concentrations of supplements and growth regulators on the growth of shoot and root. The best supplements for shoot length was 3% sucrose with 15% coconut water, for the number of leaves 0.1 mg/l NAA with 1.0 mg/l IBA or 15% coconut water with 6 g/l peptone, and for the root development 1.0 mg/l NAA with 0.5 mg/l IBA.
