Preservation method of genetic stocks of the silkworm (Bombyx mori): Conditions of ovary freezing, thawing and parthenogenetic activation.

To develop a method for long-term preservation of silkworm stocks using the ovary freezing-transplantation-parthenogenesis system, the conditions of freezing thawing and artificial parthenogenetic induction were examined. 1) Effects of cooling rate, cryoprotective agents and developmental stage of ovary on egg maturation were studied. Judging from the number of mature eggs developed from the implanted frozen thawed oocytes, ovaries of the 4th instar larvae seemed to be more suitable for cold storage than those of the 5th instar larvae. 2) Strain differences in the number of mature eggs in the adult moths developed from the frozen ovary-implanted larvae were examined. In four out of 10 strains used, the number of mature eggs decreased about 40-70% by the freezing treatment, while little effect of the treatment was observed in the remainder. 3) It was possible to confirm the findings of Kusuda et al. (1985) who reported that mature eggs developed from the transplanted frozen and thawed ovaries started to develop by artificial parthenogenetic activation.
