By the use of a synchronised cycle (60 mg MPA by intravaginal spongues and 500 IU eCG i.m.) ewes in two flocks were inseminated: 45 crossbred domestic Bovska x East Friesian ( BEF genotype) and 62 crossbred domestic Jezersko-solcavska x Romanowska (JSR genotype) sheep. Beltsville diluent (BTS) was used for preservation of semen from BEF rams and glucose-phosphate (GOE) diluent for JSR rams. Two intracervical inseminations (56 and 72 hours after removal of intravaginal spongues) were performed. Semen was stored at room temperature (20 deg C) before the first insemination over 8-hour period and over 18-hour period before the second insemination, respectively. Ewes in wich the cervix could be penetrated were inseminated with at least 60E06 motile spermatozoa suspended in 300 microL of extender. The semen was deposited within the cervix (68 % of inseminations were deep and 32 % of inseminations were mid intracervical). The lambing rate for BEF genotype was 17/45 and for JSR genotype 13/62 (chi-square= 3.68, P = 0.07) with 1.7 (BEF) and 1.8 (JSR) liveborn lambs, respectively (P = 0.75). This results suggest, that the ejaculate of rams preserved with BTS diluent is the most satisfactory method of semen storage for the long term transport at the room temperature.
