Intraspecific hybridization of Lentinus edodes by protoplast fusion.

In this study the authors used the protoplast fusion technique (PEG method) to construct 2 fusants between two different ecological types of Lentinus edodes. In fusion of L. edodes, the monokaryotic mycelia of mesothermal-megathermal large-sporophore type and mesothermal medium-sporophore type were used as parents, and clamp connection of dikaryotic mycelium was selected as natural marker. The mycelia of fusants produced a large number of clamp connections and exhibited an obvious antagonistic action against mycelia of the two parents. There were some differences in esterase isozyme and polyphenol oxidase activities between fusants and parents. The mycelium of fusants and parents. The mycelium of fusant could grow normally in PDA medium and was capable of developing fruit-body in sawdust medium.
