Deoxyribonucleic acid characterization in three mutants of Pyricularia oryzae resistant to edifenphos [India].

The parent strain and three mutants of Pyricularia oryzae (one EMS [ethyl methane sulphonate] mutant; one UV [ultra violet] mutant and one field mutant) resistant to edifenphos were chosen for the DNA characterization studies. The above mutants were found to be virulent, stable, and possess fitness of survival. Hence, in the present studies, the DNA of the three mutants of Pyricularia oryzae resistant to edifenphos is characterized. The undergraded DNA was isolated and characterized for hyperchromic effect, thermal denaturation, and base composition in terms of GC percent. The hyperchromic effect was maximum in the field mutant whereas it was least in UV mutant. The thermal denaturation was 96 degrees Celsius, 91 degrees Celsius, and 87 degrees Celsius, respectively for UV, field, EMS mutants and parent strain. The GC percent (UV absorbance method) was more than the parent strain (50.2%), in UV and field mutants (65.4%, 59.1%), but was almost the same in chemical mutant (49.2%). In the thermal denaturation method the GC percent was more in all the three mutants than the parent strain. The high percentage of GC content in the mutant strains suggest the more stable nature of the resistance. Further experiments are in progress to confirm the above results with the negatively correlated cross-resistance.
