Studies on Gossypium distant hybrid ovule culture in vitro.

Ovule culture was made on Gossypium hirsutum selfing, G. hirsutum X G. arboreum and G. hirsutum X G. thurberri. A well established culture system demonstrated that the optimum midium for increasing mature embryo rate and faster ovule growth were determined by genotype sources. The effects of auxin as well as amino acids on the growth of hybrid embryo of G. hirsutum X G. arboreum were investigated. It was found that 0.5-1 ppm IAA and 0.1 mM pro each increased the mature embryo number effectively. 0.5 ppm NAA and 0.1 mM G1n gave a higher rate of normally developed embryo. Ovule culture often produced abnormal embryos. Attempts to grow them into entire plants have so far been unsuccessful. The active carbon prevented the roots of the young plants from darkening effectively and consequently made the roots grow fast and produced more new roots.
