Lentinula edodes is a mushroom that contains many bioactive compounds important for nutrition and human health. Cultivation was performed in liquid medium containing potato extract, glucose and yeast extract at 25+/-0,3 deg C during 14 day, with shaking (140 rpm). After 10 and 14 days of cultivation the samples were obtained and prepared for the assay. The activity of enzymes was determined in the biomass of pellets and in liquid medium. The highest esterase activity of 90 (U/g), with aspirin as a substrate, was obtained in the dried biomass of pellets treated with ethanol and dried on the air. Dried biomass of pellets showed the highest peroxidase activity of 9.77 (U/g), with K4[Fe(CN)6] and hydrogen peroxide as substrates. The same sample also showed the highest peroxidase activity of 2.76 (U/g) when using 4-aminoantipyrine and phenol as substrates. Dried biomass of pellets obtained after the removal of fresh pellets from the liquid medium, washing with destilled water and air drying, showed the highest peroxidase activity 0.236 (U/g), with o-dianisidine and hydrogen peroxide as substrates. Peroxidase, MnP- and MiP-peroxidase and lacase activity in all samples was not detected with quiacol, as well as lacase with K4[Fe(CN)6] as substrate.
