Efficacy of some commercial sanitizers for devitalization and removal of bacterial biofilms

The devitalization and removal of biofilms formed by bacteria isolated from meat and ewes milk processing plants (Proteus mirabilis, Pseudomonas aeruginosa, Staphylococcus sp., Stenotrophomonas maltophilia), using eight industrial sanitizers based on benzalkonium chloride, hypochlorite, peroxides and aldehydes, were studied. Experiments were performed in polystyrene microplates. The devitalization of the biofilm was followed by the fluorescein diacetate assay, removal of the biofilm was followed by the crystal violet assay. None of the tested sanitizers was sufficiently effective with all the tested strains, either at the devitalization of the biofilm or at its removal, when used at a concentration recommended by the manufacturer and at a duration of the exposure of 10 min. Neither a prolonged exposure of 30 min, nor a three-fold increase in the concentration led to a significant improvement in the effeciency of the sanitizers. Best results at the removal of bacterial biofilm were achieved applying Ban-Kaz (Banchem, Dunajská Streda, Slovakia) or a peroxide-based sanitizer Desana-Max (Thonhauser, Perchtoldsdorf, Austria) in an ultrasonic bath, eventually with scrubbing. The results underline the need for performing mechanical cleaning together with the application of sanitizers, in order to achieve good hygiene in food processing plants
