An investigation on the effect of various buffers (Tris, Citrate and semi - skimmed milk) on ram semen motility and survival characteristics in liquid storage

Introd using an appropriate buff'er for ram semen dilution and storage is an economical and technical neccessity in sheep product ion and breeding projects. Semen was collected from 10 Mog hani rams by the use of an artificial vagina in Animal Husbandry Research Institute. Karaj, Suitable samples were pooled and divided in three protions. Each portion was diluted randomly with two volumes of Tris buffer, citrate buffer or semi-skimmed milk huffer at 30"c. Diluted semen samples were incubated either at 5 refrigerator or at 16 water bath. progress linear motility and percentage of' live spermatozoa were evaluated by light rnicroscopie. The percentage of motility of spermatozoa immediately after dilution in Tris buffer, citrate buffer and semi-skimmed milk buffer were 83.17, 80.5 and 82.8% respectively. while after 36 hours of incubation the motility percentage were 13.40 , 10.8and 11.75% respectively . The percentage of motility and the percentage of live spermatozoa were shown to be significantly (PO.OS) lower for' diluted semen samples in citrate buffer than semen Samples we re diluted in either Tris buffer or semi - skimmed milk The conclusions that the semen from Moghani rams can be diluted and stored in liquid condition at either 5c or 16°C for further use. However, its survival characteristics were reduced after 12 hours of incubation
