Micropropagation was employed for the rapid propagation of the hybrid clone Populus deltoides x Populus italica cv. He - 89/2. The initial explants were stem pieces consisting of the nodal part which were collected from hardwood cuttings, sterilized and established in vitro. Establishment of explants that formed stable shoot cultures was achieved on modified Woody Plant Medium (WPM) supplemented with 0.2mg/1 6-(gamma-gama-dimethyIlalylamino)purine. Maximum shoot proliferation occured when explants with 2-3 nodes were placed horizontally on WPM in the presence of 0.1mg/1 benzylaminopurine. Shoot growth was more vigorous in the absence of growth regulators on basal medium. Microshoots rooted by 60 percent when treated with 0.001 mg/1 alpha-Napthaleneacetic acid. Following a hardening procedure on peat : perlite mixture (1:1, v/v) the rooted microshoots grew normally in the greenhouse. The present study indicates that micropropagation can be used to achieve stable shoot cultures of recalcitrant Populus species.
