Polyhydroxybutyrate (PHB) production by Alcaligenes lactus TISTR 1403 treated with gamma radiation and 2-aminoanthracene

In the present study of A. lactus TISTR 1403 were treated with gamma radiation to 0.8 kGy before exposed to 2-aminoanthracene (2AA). When cultivated in modified PHB production medium with glucose as carbon source, A. lactus TISTR 1403 /gamma-2AA produced PHB higher than parental strains for 59.2 percent of cell dry weight. Comparison of different carbon sources as a fructose, sucrose, sucrose commercial grade (mitr phol white sugar, mitr phol natural mineral sugar and mitr phol brown sugar), maltodextrin commercial grade, cassava starch and cassava pulp hydrolysate. It was found that when the mitr phol natural mineral sugar (sucrose commercial grade) was used as carbon source, A. lactus TISTR 1403 /gamma-2AA production PHB was 2.77 (g/L), compared with controls (0.8 g/L). When cultivated on the cassava pulp hydrolysate, PHB production was similar to that found on glucose. Adjustment of initial concentration of cassava pulp hydrolysate to 30-70 percent (v/v) showed that the culture produced maximum PHB content as 50 percent (1.08 g/ L). Regarding to nitrogen sources selection, ammonium sulfate, ammonium chloride, ammonium nitrate, ammonium acetate and urea were considered. A. lactus TISTR 1403 /gamma-2AA produced of 5.25 and 2.08 (g/L) cell dry weight and PHB yield, respectively when cultivated in the presence of ammonium nitrate and 50 percent cassava pulp hydrolysate.
