Cultivation of Lentinus polychrous Lev. on cassava rhizome was studied under different conditions. Dry weight loss of cassava rhizome (percent) was used as an indicator for growth of L. polychrous Lev. Box-Behnken Design was used for experimental design with three factors: initial moisture content in cassava rhizome (X1, 30-70 percent), temperature (X2, 20-37 deg C) and cassava rhizome size (X3, 0.85-15 mm). Initial moisture content in cassava rhizome was the most significant factor influence on dry weight loss of cassava rhizome. The second order polynomial regression model obtained was fitted and found adequate, with an R sup (2) of 0.921 (p LT 0.05). Dry weight loss of cassava rhizome was maximum at 22.46 percent after cultivation of L. polychrous Lev. for 3 weeks on cassava rhizome that has the initial moisture content between 50-70 percent under the temperature of 34-37 deg C and size of cassava rhizome that was used in cultivation is 0.85-1.85 mm.
