Glucose oxidase (GOX) enzyme can generate H2O2 by the oxidation of glucose in the presence of molecular oxygen. H2O2 has the potential to enhance biotic and abiotic stress tolerance of higher plants. The gene encoding glucose oxidase is isolated from the genomic DNA of Penicillium funiculosum. The GOX gene is cloned under the control of a CaMV 35S promoter into the plant transformation vector pBI121. Potato and tobacco plants are transformed via Agrobacterrium tumefaciens-mediated procedure by selecting for kanamicyn resistance. Stable integration and expression of GO gene in the transgenic lines are ascertained by molecular and biochemical tests. Transgene integration in the transformants is confirmed by PCR for GOX gene, NOS-terminator, actin gene, VirE2 gene. Histochemical analyses for H2O2 production are studied using starch-KI medium.
