The objective of this study was to develop suitable methods for rapid in vitro shoot multiplication and rooting of two sweat cherry cultivars (Karesova, Rivan). Six proliferation MS media containing 1, 2 and 4 mg/l BAP (6-benzylaminopurine), 0.5 and 1 mg/l TDZ(thidiazuron) or lO mg/l 2iP (6-(g,g-dimethylallylamino)purine) were tested. For the two cultivars, the effect of two growth regulators in different concentrations on proliferation is shown. Multiplication rates varied depending on the genotype and medium used. The highest multiplication rate was obtained for cultivar Rivan that produced 3.1 +/- 0.1 shoots (longer than 10 mm) on MS medium containing 2 mg/l BAP. The lowest multiplication rate (1.2 +/- 0.1) was obtained for cultivar Rivan on MS medium containing 10 mg/l 2iP. three auxins IBA (indole-3-butyric acid), IAA (indole-3-acetic acid) and NAA (1-naphthaleneacetic acid) in concentration 1 mg/l were tested for rooting of cultivar Rivan. The best result of rooting (40 %) was obtained for auxin IAA.
